Ensysce Biosciences Puts Psychedelic Pain Therapy in the Spotlight Ahead of FDA Hearing

Ensysce Biosciences will participate in the U.S. Food and Drug Administration’s September 14 public hearing on the potential future therapeutic use of psychedelic drugs, as the company develops a psychedelic-derived treatment for chronic pain.

The U.S. biotechnology company (NASDAQ: ENSC) is scheduled to join the FDA hearing alongside a series of investor and industry events in September.

The FDA’s hearing, titled Considerations for Potential Future Therapeutic Use of Psychedelic Drugs, will take place on September 14 at the agency’s White Oak Campus in Silver Spring, Maryland, with online participation also available.

The hearing is intended to gather views on the scientific, regulatory and practical considerations involved in the potential therapeutic use of psychedelic drug products in supervised and supportive settings.

Ensysce’s interest in the discussion comes from its recent acquisition of Cy Biopharma, which added the clinical-stage psychedelic-derived candidate CY200 to its pipeline.

CY200 is being developed for Complex Regional Pain Syndrome (CRPS) Type 1, a severe chronic pain condition for which there is currently no FDA-approved treatment. The programme has received FDA Orphan Drug Designation.

Developing a psychedelic-derived treatment for pain

Ensysce describes CY200 as a neuroplastogenic therapy and is investigating its potential to address the neurological processes associated with CRPS rather than focusing solely on symptom relief.

The company says CY200 uses multi-tryptamine pharmacology to target central sensitisation, a process associated with persistent pain in CRPS.

The approach puts Ensysce in a part of the psychedelic medicine sector that remains relatively small. Much of the industry’s clinical development to date has focused on psychiatric conditions including depression, PTSD and substance use disorders, while research into psychedelic and psychedelic-derived treatments for physical pain is still developing.

Ensysce completed its acquisition of Cy Biopharma in August. The transaction was accompanied by financing of up to approximately US$77 million, including an initial US$21.5 million private placement, US$17.1 million in Cy Biopharma cash and a potential additional US$38.6 million linked to a clinical milestone.

The company says the funding will support development of CY200 through a randomised Phase 2 proof-of-concept study and towards registrational development.

CY200 is being developed alongside Ensysce’s existing pain and opioid-safety programmes.

The company is also developing PF614-MPAR, an opioid candidate using its MPAR technology, which is designed to reduce the risk associated with oral overdose. Ensysce announced on August 31 that the final phase of the PF614-MPAR-102 clinical study had been completed.

What the FDA hearing is about

The September 14 hearing is not an approval process for psychedelic medicines.

Instead, the FDA is seeking public and expert input on the issues that may need to be addressed if psychedelic drug products move further into therapeutic development and potential clinical use.

Those issues could include clinical trial design, safety, manufacturing, dosing, psychological support and the regulatory requirements associated with administering psychedelic medicines in supervised settings.

The FDA is also accepting written comments through the Federal Register process until October 5, 2026.

For companies developing psychedelic medicines, the regulatory approach taken by the FDA will be relevant to how future programmes are designed and evaluated.

Ensysce’s participation gives the company an opportunity to contribute to that discussion, but it does not represent an endorsement of CY200 by the FDA.

Ensysce’s September schedule

The FDA hearing is one of several events on Ensysce’s calendar this month.

The company is scheduled to present at the 2026 Cantor Global Healthcare Conference in New York on September 10 and participate in the H.C. Wainwright 28th Annual Global Investment Conference from September 14 to 16.

It will also participate in the Life Sciences Product Launch Summit in Boston on September 22 and the 20th Annual Pain Therapeutics Summit in San Francisco on September 28 and 29.

The events come shortly after Ensysce expanded its pipeline through the Cy Biopharma acquisition.

CY200 now gives the company a psychedelic-derived programme alongside its existing work in pain and opioid safety.

The next major test for the programme will be clinical. Ensysce will need to show that CY200’s proposed mechanism translates into meaningful improvements for people with CRPS.

That evidence will ultimately matter more than the company’s presence at industry or regulatory events.

For now, Ensysce is one of a small number of biotechnology companies exploring psychedelic-derived compounds outside the traditional focus on mental health, with its CY200 programme aimed at a difficult and poorly served chronic pain condition.
