Compass Pathways has reported positive six-month results from its second Phase 3 clinical trial evaluating COMP360 psilocybin treatment for treatment-resistant depression (TRD), reinforcing the durability of the company’s late-stage clinical programme as it progresses towards a U.S. regulatory submission.
The COMP006 trial found that a single 25 mg dose of COMP360, administered alongside structured psychological support, produced rapid improvements in depressive symptoms that were maintained through 26 weeks. According to the company, the study also demonstrated a safety profile consistent with previous COMP360 trials, with no new safety signals identified.
The results represent another significant milestone for Compass Pathways, which is seeking to become the first company to secure U.S. Food and Drug Administration (FDA) approval for a prescription psilocybin therapy.
Treatment-resistant depression affects patients who have not responded adequately to multiple antidepressant therapies and remains one of the most challenging psychiatric conditions to treat. Current treatment options often require ongoing medication and can take several weeks before therapeutic effects become apparent.
Compass has developed COMP360 as a pharmaceutical-grade synthetic psilocybin treatment administered in a controlled clinical setting alongside psychological support. The company has consistently positioned the programme as a single-dose intervention designed to deliver durable antidepressant effects rather than requiring frequent dosing.
According to Compass, patients in the COMP006 study experienced antidepressant effects shortly after treatment, with clinically meaningful improvements maintained throughout the six-month follow-up period. The company said most treatment-emergent adverse events were mild to moderate in severity and resolved without long-term complications.
The findings build on the company’s earlier positive Phase 3 results and strengthen the evidence supporting COMP360 as it advances towards regulatory review.
Compass has already commenced a rolling New Drug Application (NDA) submission with the FDA and expects to complete the filing during the fourth quarter of 2026. If accepted and approved, the company continues to target a potential U.S. commercial launch during the first half of 2027.
The announcement is likely to be viewed as one of the most significant clinical developments for the psychedelic medicines sector this year. Successfully reproducing positive efficacy and safety findings across two pivotal Phase 3 studies reduces a major source of late-stage clinical development risk and shifts investor attention towards the regulatory review process and commercial preparation.
Should COMP360 receive FDA approval, it would become the first federally approved psilocybin-based treatment in the United States, marking a major milestone for the emerging psychedelic medicines industry.
Approval, however, would represent only the beginning of commercial adoption. Questions surrounding reimbursement, treatment delivery, therapist capacity and healthcare system integration are expected to play a significant role in determining how quickly psychedelic-assisted therapies become accessible to patients following any regulatory decision.
With two positive Phase 3 studies now completed and its NDA submission approaching completion, Compass Pathways has moved into the final stages of a clinical development programme that has spanned more than a decade. The industry’s attention will now turn to the FDA review process, which could shape the future trajectory of psychedelic medicine in the United States.
